Why “lifecycle” is the modern default
Traditional thinking treats method validation as a one-time gate: develop, validate, file, done. The modern view treats an analytical procedure as a controlled system that must stay fit-for-purpose through design, qualification, and ongoing verification.
That lifecycle framing is embedded in USP <1220> Analytical Procedure Life Cycle, which uses an Analytical Target Profile (ATP) plus three lifecycle stages, and it became official in May 2022. U.S. Food and Drug Administration+1

This article turns the checklist into a practical way to plan and document method work without overengineering.
1) Draft a one‑page ATP before starting development
If you skip the ATP, method development becomes a cycle of “try conditions until it looks okay,” and validation becomes a box-checking exercise. The ATP forces the team to agree on what the method must accomplish (and how you’ll know it’s good enough) before anyone touches a column, detector, or integration parameter.
USP <1220> explicitly positions the lifecycle around the ATP and the three stages; the ATP defines the required performance characteristics for the procedure. USPNF+1

2) Plan a small, focused DoE to map critical factors
A “modern” method program doesn’t rely on one-factor-at-a-time tweaks. It uses focused experimental design to identify critical method parameters and understand how they affect critical method attributes (e.g., resolution, bias, precision, detection capability).

How to keep DoE “small and useful” (not academic)

The biggest gap between “validated” and “works in production” is that validation often happens under ideal conditions, while routine testing includes:

“Mirror real use cases” — practical moves

Lifecycle doesn’t end at validation approval. If Stage 3 isn’t planned, teams discover method fragility only after OOS events, failed transfers, or trends that should have been visible earlier.
A follow-up review is the simplest way to convert “we validated” into “we operate in control.”
